Summary A study was made of substance P-like-immunoreactive nerves within the small intestine of platypus, an Australian prototherian mammal. Both immunoreactive nerve cell bodies and fibres were present. No immunoreactive fibres were found in mesenteric nerves or on intramural blood vessels, suggesting that extrinsic sensory neurons containing substance P do not innervate platypus ileum. This was further supported by the result of in vitro experiments. Although applied substance P (10-9 M-3 × 10-8 M) caused contraction of the longitudinal muscle, neither mesenteric nerve stimulation nor application of capsaicin caused contractions.  相似文献

The intermediate filament protein vimentin was phosphorylated with cAMP-dependent protein kinase under conditions that induce filament disassembly. Digestion of phosphorylated vimentin with lysine-specific endoprotease and subsequent tryptic peptide mapping indicated that a 12 kDa N-terminal fragment contained all the phosphorylation sites found in the intact molecule. Analysis of cyanogen bromide digests indicated that two phosphorylated peptides were produced, with the major 32P-labeled species representing amino acid position 14-72, and a minor 32P-labeled peptide representing amino acid positions 1-13. These results demonstrate that phosphorylation of sites within the N-terminal head domain of vimentin are associated with phosphorylation induced filament disassembly.  相似文献

Endosome fractions were prepared from rat liver homogenates, and antibodies were raised in rabbits against the integral membrane proteins. Immunofluorescent studies showed that these antibodies identified primarily intracellular structures in liver sections, isolated hepatocytes and HepG-2 cells. Immunoelectron microscopy using protein A-gold confirmed that endocytic multivesicular structures, especially those located at the biliary pole of the hepatocyte, were labeled. Biochemical analysis showed that approximately 12 endosome antigens were present. A major 43 kDa glycosylated antigen corresponded to the asialoglycoprotein receptor subunit. A further antigen identified in endosomes was a 115 kDa polypeptide pI 4.3 previously identified as a major calmodulin-binding protein. The antigens identified in rat liver endosomes were different to those previously shown by other studies to be present in the Golgi apparatus and lysosomes.  相似文献

Evidence for the non-indolic nature of the new citrus auxinis presented on the basis of fluorometric properties, thin-layerchromatography, Ehrlich's colour reaction, paper electrophoresis,and the infra-red spectra determinations. Citrus auxin had alower Rf in TLC than IAA, did not give the typical indole reactionwith Ehrlich's reagent, and behaved differently in electrophoresis.The infra-red spectra also provided preliminary informationconcerning chemical structure. The hypothesis that indolic compoundsconstitute the only natural auxins in higher plants should berevised in view of this evidence that a non-indole auxin existsin higher plants.  相似文献
